“Of cortex and cognition in Parkinson's disease”

Cognitive dysfunction is a source of major disability in Parkinson's disease. The cause of PD-related cognitive dysfunction is unknown. We have found that PD patients have increased variability during interval timing, or estimating intervals of several seconds. This variability is correlated with attenuated ~ 4 Hz cortical rhythms. Strikingly, our work suggests that rodents have similar 4 Hz cortical rhythms during interval timing, and these rhythms require cortical dopamine in rodent PD models. We have found that these 4 Hz brain rhythms synchronize cortical networks in service of cognitive control. We discuss strategies to exploit 4 Hz synchrony to compensate for dopamine-dependent deficits in cognitive function.
